He was actually criticising the mystical thinking of the Sith, not the scientific reality of midichlorians.
Wasn't he more criticizing the scale? Like, a sith warrior is great, but he can't really compete with the power of the Death Star when it comes to sheer scale. Just ask Alderaan.

Vader was right. I didn’t know the powah of the dark side.
Force drain is one thing where he always has to be constantly feeding to survive. The Death Star, especially 2 with it's 1 min recharge time, if it had been finished could jump to any systen fire at any planet and vaporize it. Then it could wait a few minutes with its incredible defenses or jump away within a few seconds.
The death star is terrifying when the Empire is actually competent.
